Genome-scale engineering of Saccharomyces cerevisiae with single-nucleotide precision.

CRISPR–Cas9- and homology-directed-repair (HDR)-assisted genome-scale engineering (CHAnGE) uses plasmids containing HR arms to frameshift or point-mutate precise locations, which are cleaved by Cas9 using a guide RNA expressed by the same plasmid. Plasmid libraries are made by cloning libraries of long oligonucleotides synthesised on microarrays.
